If you're arriving early in the day, my advice would be to walk from the station to Fässla, though I'm not a fan of theirs, but I think they're worthy of a visit. Then across the street to Spezial and have an "U", if it's on, which it's not always, and lunch.

Then to the centre, taking in the sights along the way, up to Klosterbräu. If the weather's nice, way up to the Spezial Keller for more of their U and Rauchbier (open from 15.00). Then back down to Schlenkerla. If you like typical German food, have the specialty there, a Bamberger Zwiebel, and as much Rauchbier as you like.

If you like to do a lot of walking, you could then trek out to Keesmann and Mahr's, but IMO they're a bit of a let-down after Schlenkerla. Buses can get you around too.
